Camptothecin is a naturally occurring alkaloid derived from the plant Camptotheca acuminata that was identified in an anticancer drug discovery screen in the 1960s. Camptothecin forms a stable ternary complex, preventing normal DNA re-ligation, and causes the complex to collide with the replication fork, leading to a DNA double-strand break and cytotoxicity.
Camptothecins are mainly used in lung, breast, and brain cancer. However, topotecan is approved by the USFDA for the treatment of lung and ovarian cancer and irinotecan is approved for the treatment of colon cancer.
